    Mdm2 is overexpressed in many human cancers, especially in human breast cancer. Its overexpression affects the process of tumorigenesis. So detecting the variation of Mdm2 mRNA expression can monitor the tumor progression. However, traditional methods can't be applied to detect the variation of Mdm2 mRNA in situ. In this study, we designed a new strategy for detecting the variation of Mdm2 mRNA expression with gold nanoparticle, in other words, a special hairpin deoxyribonucleic acid (DNA) for Mdm2 mRNA to functionalize gold nanoparticles, which served as a beacon for detecting the variation of Mdm2 mRNA expression. A-quenching efficiency up to 90% was achieved. The beacon we designed in this study can monitor the precise variation of Mdm2 mRNA expression in situ. The strategy reported in this study is a promising approach for the measurement of mRNA expression in living cells. Moreover, it has a great potential in the study of drug screening and discovery.
